Just one way? What sort of time? Are you happy to be tied down to one particular service? Railcards?

If I were you I wouldn't start from there... ;)
Winchester-Basingstoke shut
Wolverhampton-Stafford-Stoke/Crewe shut

However, XC are running hourly Bournemouth-Manchester with two diversions via Andover and Nuneaton taking 6 hours.

For fares, it depends on when you set off and if/when you are coming back.
Off Peak Single is £119.40 (£120.90 via London).

NRE says there's a £24.50 ticket valid on the 18:59 from Bournemouth to London, and you can change at Clapham Junction and Watford Junction, reaching Manchester at 2:07 the next morning. Any good?

That ticket seems to be £18.00 (via Havant/Waterloo and Euston) on the XC site, and on certain through trains there are advances at £58.10.

Best public transport option would appear to be:
Bournemouth-Southampton parkway(SWT)
(one evening flight) and flybe sou to man
Do Flybe offer Flybe and Rail connections tickets?

If you want to avoid going via London, then it looks like there's only one coach option, National Express service NX333 departs Bournemouth 07:45 and arrives in Manchester at 17:45 and costs £59. All other coach options that day show one change required, without going through them I'd assume that'd be via London Victoria but wouldn't be surprised if a couple were via Heathrow.
