If splitting day Reading to London (with Travelcard) tickets at Twyford, the service obviously has to stop at Twyford to be valid.

With a Reading to Twyford season, I think Twyford to London day tickets are valid without stopping at Twyford. However, does this include Twyford tickets with the not via Reading restriction?

Yes, a non-stop split at Twyford would be permitted with that combination of tickets. The Twyford-London ticket being routed "not via Reading" is absolutely fine as the Reading-Twyford season covers that part of the journey.
