Would a Swindon - Cardiff single/return that states 'valid only via Newport' be valid via Gloucester?

Yes, there was regular diversions via Gloucester during the electrification, and during Severn tunnel closures.

Realistically the direct line, Gloucester and via Bristol would all be valid

The only alternative (not via Newport) is a really indirect routing via Heart of Wales line which you would be mad to use anyway.
