Class 221 - units 221142 and 221143

As I understand, the last four Class 221 units to be built for Virgin Trains, units 221141 to 221144, were originally four-cars (the rest were five-cars).

But around 2010, units 221142 and 221143 were extended to five-cars by taking vehicles off unit 221144.
Unit 221142 received 60994 (an ex MSL(A) vehicle), while 221143 received 60794 (an ex MSRMB vehicle).

As a result, does that mean that the unit formation for units 221142 and 221143 are different from the rest of the Class 221 fleet?

It did mean that 221143 had two shop vehicles as you note.

However, since Crosscountry resurrected unit 221144, there's been some reformation of 221142 and 143 so that they're now formed:

221142 60392-60992-60986*-60792-60492
221143 60393-60993-60994**-60793-60493

*Motor Standard vehicle formerly in 221136.
**Motor Standard vehicle originally in 221144, then latterly 221142.

Both units therefore have two 609xx MSL(A)s, whereas all other 5-car 221s each have one 608xx (MSL(B)) and one 609xx (MSL(A)).
